低温保存过程中老鼠成骨细胞胞内冰的研究

摘要:
低温保存过程中,胞内冰是造成细胞损伤的主要因素之一,运用实验与模型相结合的手段,研究细胞胞内冰的生成规律,可以为细胞低温保存方案的设计提供可靠的依据。应用低温显微镜系统,结合胞内冰生成模型,系统地研究了老鼠成骨细胞在低温保存过程中的胞内冰生成规律。在首先获得水渗透参数及胞内冰成核参数的基础上,应用数学模型预测了多种降温速率下老鼠成骨细胞的胞内冰生成率,结果表明,实验数据与模型计算能够很好的吻合。

Intracellular Ice Formation of Mouse Osteoblasts During Cryopreservation

Abstract:
Intracellular ice formation(IIF)is one of the factors that causes cell damage during cryopreservation pro-cess.The study of IIF will provide fundamental information for the development of appropriate cryopreservation proto-cols.The possibility of IIF at various cooling rates for mouse osteoblasts was studied by using a cryomicoscopy sys-tem.The water transportation and ice nucleation parameters were used in the coupled water transport.ice nucleation model to predict the probability of IIF over a wide range of cooling rates.The calculated probability of IIF was in good agreement with the experimental data,suggesting the validity of the predicted parameters.
